First-generation antipsychotics (also known as conventional or classical antipsychotics) are dopamine antagonists. They bind to dopamine receptors to block dopamine neurotransmission. These drugs include chlorpromazine, fluphenazine and haloperidol and are associated with a high risk of side effects due to their actions on other transmitter systems in the brain and periphery, eg cholinergic, adrenergic and histaminergic receptors, leading to changes in cardiovascular function and sedation, etc. |
