First-generation antidepressants include the tricyclics (TCAs) and the monoamine oxidase inhibitors (MAOIs). Both classes of drugs work to reduce the symptoms of depression by increasing the concentrations of noradrenaline and serotonin at the synapse, so facilitating neurotransmission by prolonging the effects of these Neurotransmitters. They are often associated with a relatively high level of side effects.
